This essay is an attempt to point up the gap between, on the one hand, the methods currently available to the biologist in the laboratory and, on the other, the kind of data that he or she would need in order to characterise genetically engineered proteins of topical biological interest in such a way as to make use of the techniques of protein engineering.

Deep brain stimulation of the subthalamic nucleus is a clinically effective tool for treating medically refractory Parkinson’s disease, but its neural mechanisms remain debated. Previous work has demonstrated that STN DBS results in evoked potentials in the primary motor cortex, suggesting that modulation of cortical physiology may be involved in its therapeutic effects. Due to technical challenges presented by high-amplitude DBS artifacts, these EPs are often measured in response to low-frequency stimulation, which is generally ineffective at PD symptom management. This (...) study aims to characterize STN-to-cortex EPs seen during clinically relevant high-frequency STN DBS for PD. Intraoperatively, we applied STN DBS to 6 PD patients while recording electrocorticography from an electrode strip over the ipsilateral central sulcus. Using recently published techniques, we removed large stimulation artifacts to enable quantification of STN-to-cortex EPs. Two cortical EPs were observed – one synchronized with DBS onset and persisting during ongoing stimulation, and one immediately following DBS offset, here termed the “start” and the “end” EPs respectively. The start EP is, to our knowledge, the first long-latency cortical EP reported during ongoing high-frequency DBS. The start and end EPs differ in magnitude and latency, and the end, but not the start, EP magnitude has a significant relationship to ongoing high gamma power during the EP. These contrasts may suggest mechanistic or circuit differences in EP production during the two time periods. This represents a potential framework for relating DBS clinical efficacy to the effects of a variety of stimulation parameters on EPs. (shrink)

We examine three critical aspects of Popper’s formulation of the ‘ Logic of Scientific Discovery ’—evidence, content and degree of corroboration—and place these concepts in the context of the Tree of Life (ToL) problem with particular reference to molecular systematics. Content, in the sense discussed by Popper, refers to the breadth and scope of existence that a hypothesis purports to explain. Content, in conjunction with the amount of available and relevant evidence, determines the testability, or potential degree of corroboration, of (...) a statement; content distinguishes scientific hypotheses from metaphysical assertions. Degree of corroboration refers to the relative and tentative confidence assigned to one hypothesis over another, based upon the performance of each under critical tests. Here we suggest that systematists attempt to maximize content and evidence to increase the potential degree of corroboration in all phylogenetic endeavors. Discussion of this “total evidence” approach leads to several interesting conclusions about generating ToL hypotheses. (shrink)

During the Covid‐19 pandemic, as resources dwindled, clinicians, health care institutions, and policymakers have expressed concern about potential legal liability for following crisis standards of care (CSC) plans. Although there is no robust empirical research to demonstrate that liability protections actually influence physician behavior, we argue that limited liability protections for health care professionals who follow established CSC plans may instead be justified by reliance on the principle of reciprocity. Expecting physicians to do something they know will harm their patients (...) causes moral distress and suffering that may leave lasting scars. Limited liability shields are both appropriate and proportionate to the risk physicians are being asked to take in such circumstances. Under certain narrow circumstances, it remains unclear that the standard of care is sufficiently flexible to protect physicians from liability. Given this uncertainty, the likelihood that physicians would be sued for such an act, and their desire for such immunity, this limited protection is morally legitimate. (shrink)

‘Teachers expect parents to teach you. Parents expect teachers to teach you. So actually you learn nothing and nobody wants to talk about it’. This quote from this research study is an adolescent girls’ cry for liberation from the silence related to sexuality because of the general reluctance of adults to talk to them about it. Given the growing concerns raised about the sexual and reproductive health of adolescents in South Africa, the aim of this study was to conduct research (...) with adolescent girls as subjects in order for girls’ voices to enhance adult researchers’ understanding of children’s need for support and guidance in the context of sexuality education. The purposively selected sample included 75 participants from three diverse high schools in the Western Cape. The research was conducted using a phenomenological approach that values the lived experiences of participants as significant in contributing to the knowledge on adolescent sexuality. A qualitative interpretative research design was applied to collect the data. This article argues that adults cannot help adolescents in their sexual emancipation – to be free – if they themselves are not free. If they were free, they would have been able and willing to engage with adolescents in every aspect, which includes their sexuality. Including adolescent girls as partners in transforming sexuality education is presented as a core principle for the sexual emancipation of both adolescent girls and adults. (shrink)

Calcium storage is one of the functions commonly attributed to the endoplasmic reticulum (ER) in non‐muscle cells. Several recent studies have added support to this concept. Analysis of reticuloplasm, the luminal ER content, has shown that it contains several proteins (reticuloplasmins) which are prospective calcium storage proteins. One of these, calreticulin, is also present in the sarcoplasmic reticulum (SR). In sea urchin eggs, a calsequestrin‐like protein has been clearly localised to the ER. The recent demonstration that the IP3 receptor, which (...) has similarities with the calcium release channel in the SR is also localised in the ER membrane suggests that calcium stored in the ER is important for intracellular signalling. The alternative view, that the physiologically important calcium store is a specialised organelle, the calciosome, is not supported by these observations. Recent evidence also suggests that ER calcium might be important in ER structure and in the retention of the luminal ER proteins. (shrink)

It is often assumed that engaging in a one‐on‐one dialogue with a tutor is more effective than listening to a lecture or reading a text. Although earlier experiments have not always supported this hypothesis, this may be due in part to allowing the tutors to cover different content than the noninteractive instruction. In 7 experiments, we tested the interaction hypothesis under the constraint that (a) all students covered the same content during instruction, (b) the task domain was qualitative physics, (c) (...) the instruction was in natural language as opposed to mathematical or other formal languages, and (d) the instruction conformed with a widely observed pattern in human tutoring: Graesser, Person, and Magliano's 5‐step frame. In the experiments, we compared 2 kinds of human tutoring (spoken and computer mediated) with 2 kinds of natural‐language‐based computer tutoring (Why2‐Atlas and Why2‐AutoTutor) and 3 control conditions that involved studying texts. The results depended on whether the students' preparation matched the content of the instruction. When novices (students who had not taken college physics) studied content that was written for intermediates (students who had taken college physics), then tutorial dialogue was reliably more beneficial than less interactive instruction, with large effect sizes. When novices studied material written for novices or intermediates studied material written for intermediates, then tutorial dialogue was not reliably more effective than the text‐based control conditions. The target article misrepresents the foundations of integrated information theory and ignores many essential publications. It, thus, falls to this lead commentary to outline the axioms and postulates of IIT and correct major misconceptions. The commentary also explains why IIT starts from phenomenology and why it predicts that only select physical substrates can support consciousness. Finally, it highlights that IIT's account of experience – a cause–effect structure quantified by integrated information – has nothing to do with “information transfer.”.

Die berühmte Philosophin Hannah Arendt, deren Name heute in Deutschland Schulen, Straße, Institute und auch ICE-Züge tragen, wurde 1906 in Hannover als Kind einer jüdischen Familie geboren. Sie las Philosophie in Heidelberg bei Martin Heidegger, mit dem sie auch eine heimliche Liebesbeziehung verband. Nach der Flucht vor den Nazis 1933 über Frankreich in die USA wurde Arendt mit einem Schlag berühmt, als sie 1963 ihr Buch "Eichmann in Jerusalem" veröffentlichte. Sie machte sich über die Grundlagen politischen Handelns vor allem anhand (...) totalitärer Systeme Gedanken und hielt dabei stets betont Abstand zu allen Denkschulen der Linken oder Rechten.Diese Unabhängigkeit, die sie selbst gerne als "Denken ohne Geländer" umschrieb, ist laut Sontheimer eine der wichtigsten Ursachen für ein nach dem Ende des Kalten Krieges wieder stark gestiegenes Interesse an Arendts Texten. Anhand des Lebenslaufs der Philosophin zeichnet Sontheimer ihre Denkansätze nach. "Das Hohelied der Freundschaft" überschreibt er ein sehr eindrucksvolles Kapitel über die intensiven, zum Teil über viele Jahrzehnte über hunderte von Briefen gepflegten Beziehungen der Autorin zu Zeitgenossen wie dem jüdischen Gelehrten Gershom Scholem, ihrem deutschen Lehrer und Kollegen Karl Jaspers und der US-Schriftstellerin Mary McCarthy. This paper considers the place of reasons in the metaphysics of epistemic normativity and defends a middle ground between two popular extremes in the literature. Against members of the ‘reasons first’ movement, we argue that reasons are not the sole fundamental constituents of epistemic normativity. We suggest instead that the virtue-theoretic property of competence is the key building block. To support this approach, we note that reasons must be possessed to play a role in the analysis of central epistemically normative (...) properties, and argue that the relation of possession must be analyzed in terms of competence. But while we diverge with reasons-firsters on this score, we also distance ourselves from those who deny reasons any important role in epistemology. For we maintain that possessed reasons do help to ground deontic facts in the epistemic domain (e.g., facts about what one epistemically ought to believe, may believe, or is justified in believing). Indeed, we present an argument that the possession of sufficient epistemic reasons is necessary and sufficient for propositional justification, and that proper basing on such reasons yields doxastic justification. But since possession and proper basing are themselves grounded in competence, reasons are not the end of the explanatory road: competence enables them to do their work, putting them—and us—in the middle. We argue for a novel view of suspending judgment properly--i.e., suspending judgment in an ex post justified way. In so doing we argue for a Kantian virtue-theoretic view of epistemic normativity and against teleological virtue-theoretic accounts.

On April 20, 1999, Eric Harris and Dylan Klebold shot and killed fifteen people at their high school in Columbine, Colorado. National media dubbed the event a “school shooting.” The term grimly expanded over the next several years to include similar events at army bases, movie theaters, churches, and nightclubs. Today, we commonly use the categories “mass shooter” and “mass shooting” to organize and classify information about gun violence. I will argue that neither category is an effective tool for reducing (...) gun violence and use empirical data to show how these categories perpetuate a moral panic that harms already vulnerable demographics. I conclude that we should instead favor a narrower description of individuals and events, (e.g., “X shot Y people at Z”) because we can talk about all of the relevant cases without contributing the undue harms. Resumo O fundamento teórico deste artigo compreende uma análise teológica, com especial interesse pelo diálogo com a sociologia e com a antropologia. Investiga as relações de dominação, com enfoque específico nas relações de gênero que se engendram a partir dos ritos religiosos ou profanos, Objetiva-se verificar a importância dos ritos para a construção da realidade social, sobretudo das noções de gênero e das relações que delas advêm; expor com embasamento antropológico e sociológico esta premissa, visando contribuir para a discussão da (...) função social da religiosidade como construtora da realidade social; oferecer uma compreensão dos ritos enquanto espaços nos quais podem surgir relações mais igualitárias entre homens e mulheres. Através da pesquisa bibliográfica, procurar-se-á na sociologia de Pierre Bourdieu conceituar o poder simbólico e compreender de que maneira ele é elemento que constrói noções de gênero; posteriormente, serão averiguados em obras da área da antropologia, conceitos de rito, estabelecendo sua ligação com o poder simbólico e seu possível potencial construtor da realidade social e das noções de gênero que orientam as relações entre mulheres e homens. A conclusão buscará apontar de que maneira podem os ritos se tornar espaços para a articulação de um novo modo de relações de gênero. Palavras-chave: Ritos, Sociedade, Gênero.The theoretical basis of this article consists of a theological analysis, with emphasis on a dialogue with sociology and anthropology. It investigates dominance relationships, focusing particularly on gender relationships engendered by religious or profane rites. The objective is to verify the importance of rites for the construction of social reality, above all of gender notions and the relationships that result from them; to expose this premise on an anthropological and sociological basis, seeking to contribute to the discussion of the social function of religiosity as builder of social reality; and to provide an understanding of rites as spaces where more egalitarian relationships between men and women can take place. Through bibliographical research, especially into Pierre Bourdieu's sociology, the text tries to describe symbolic power and to understand how this element builds gender notions. Then, it verifies concepts of rite in anthropological texts, establishing its connection with symbolic power and its potential to construct social reality and gender notions that guide the relationships between women and men. The conclusion tries to indicate how rites can become spaces for the articulation of a new mode of gender relationships. Key words: Rites, Society, Gender. (shrink)
